for indirect processing of samples in closed vessels

By direct immersion of tips (microtips) during sonification of toxic, pathogenic or radioactive samples contamination and aerosols may occur. For preventing such unpopular effects an indirect sonification is adviseable.

By using the "cup horn High Intensity" samples can be indirectly sonified, to say without immersion of the tip into the sample.

Also samples with volumes below 100 µl can be sonified with high power, what is not possible by immersion of a tip (microtip or needle tip) and transmitting sufficient ultrasonic power into the solution. By using the "CUP HORN High Intensity" smallest samples - without any minimum volume limit - can be sonified.

Due to the ultrasound generated heat sonification of smallest samples often causes problems. The coolant flow system of the "CUP HORN High Intensity" combined with a cooling device (e.g. FRYKAmultistar) provides optimal temperatur control of the sample. The "bath"-liquid is ultrasound transmitter and cooling media at the same time. While recycling through a cooling device, constant temperature of the sample can be kept.

In stead of the disruptor horn the "CUP HORN High Intensity" is mounted to the converter, which is turned 180° and fixed at a lab-stand. After connected to the cooling circulation the sample is immersed quasi into a high intensity ultrasonic bath and is sonified indirectly through the vessel’s wall.

Due to the parabolic design of the vibrating surface higher ultrasonic intensities are available than in cup horn systems with planar surface. (e.g. BRANSON CUP HORN 2" Dia. EDP 101-147-047 or 3" Dia. EDP 101-147-048). This parabolic design effects a strong concentration of ultrasonic power in the center of the parabolic area.

Because it is possible to sonify even smallest samples with the "CUP HORN High Intensity" at max. power setting "10" of the BRANSON SONIFIERÒ a compensation of lower power density of all indirect systems (compared with direct immersion horns) is made. The occuring loss is based on the significant more volume to bring into vibration (complete "bath" content) by lower available amplitude of the vibrating surface and also by possible absorption of vibration of the vessel wall.

Therefore the "CUP HORN High Intensity" cannot be applied to disrupt all biological material. By treatment of extremely durable microorganisms (e.g. some yeast, chlorella algaes...) the available power density of the "CUP HORN High Intensity" might be too low for sufficient yield. In some cases a significant increase of sonification time can compensate this lower intensity. While using a cooling device no heat problems will occure even by longer treatment of smallest volumes. Also the change of vessel can improve sonification results by lower absorbens rate of the vessel material
